Abstract

*Streptococcus pneumoniae* is a major cause of pneumonia, meningitis and other invasive diseases leading to morbidity and mortality among piglets from 1 to 4 months of age, as well as sows. Inaccurate identification of *S. pneumoniae* hides an accurate assessment of the severity of the disease and may delay the choice of treatment. This is a common problem most often faced by large pig farms. Conflicting detection methods often delay early identification and confirmation of *S. pneumoniae*.

Available data on the severity of the disease and the prevalence of *S. pneumoniae* have not been reliably assessed, as have the risk factors currently facing pig farming, which in turn limits the use of available pneumococcal vaccines. In this article, we review and compare methods: PCR and LAMP in real time.

Methods such as culture or antigen detection have traditionally been used for microbiological diagnosis, but they are time-consuming and insufficiently sensitive. The PCR method has reduced the time of diagnosis, but requires expensive equipment. Thus, new isothermal methods are emerging, one of which is loop-mediated isothermal amplification (LAMP).

The purpose of the study the aim was to compare the methods of real-time isothermal loop amplification (LAMP) and polymerase chain reaction with real-time fluorescence detection (PCR) for the diagnosis of Streptococcus pneumoniae in pigs. The results obtained during laboratory diagnostic studies of the biomaterial from pigs received at the Kropotkin Regional Laboratory over the past 4 years (from 2019 to 2022) indicate a high detection rate of S. pneumoniae (from 33 to 65%). Comparing the methods of PCR and LAMP, it can be noted that the isothermal loop amplification reaction time takes from 5 to 45 minutes, which allows you to conduct the same study as PCR, but much faster, and LAMP has a high specificity of 4-6 primers, and PCR only 2.
