EVALUATION OF ALLERGENIC PROPERTIES OF THE DRUG DON-1 IN THE GENERAL DESIGN OF PRECLINICAL TOXICOLOGICAL STUDIES

The development of the veterinary pharmaceutical market under the sanctions policy and the shortage of domestic drugs has recently become one of the key sectoral priorities of the state policy of the Russian Federation. In this case, with the emergence of new means of prevention and therapy of animal diseases, there is a need to develop a program of preclinical studies, including a thoughtful and justified assessment of all possible toxic or side effects of a medicinal substance, taking into account its specific action. At the same time, the mandatory design of studies includes an assessment of the allergenic properties of new drugs in laboratory animals, characterizing hypersensitivity and the development of adverse reactions.
The problem of drug allergy is also that it is often impossible to accurately predict allergic reactions based on the structure, molecular weight and mechanisms of action of a drug or chemical compound. Therefore, the assessment of the allergenic hazard of new drug compounds and complex products based on them should be carried out based on the conditions of intended use and the possible damaging effect on the skin and mucous membranes of experimental animals.
The article presents the results of evaluating the allergenic properties of the drug DON-1 on the example of express sensitization of the skin of guinea pigs and revealing its irritating effect on the mucous membranes of the eyes of rabbits. It has been determined that during permissive epicutaneous sensitizing skin testing, the sample of the DON-1 drug does not have a local irritant effect, while the evaluation of the conjunctival test indicators revealed a pronounced potential allergenic activity of the drug, enhanced by the toxic effect of its constituent components.
